چکیده

Tetrahydrocannabinol (THC) derivatives are used clinically as analgesic, anti-inflammatory, appetite stimulant, anti-emetic and anti-tumor cannabinoids. THC and its related compounds are at present obtained by extraction from intact Cannabis plants or chemical synthesis, but plant cell cultures may be an alternative source of production. In the present study, hairy root cultures of C. sativa (Cannabaceae) were induced by incubation of aseptically grown callus culture with solid B5 medium supplemented with 4 mg/l naphthaleneacetic acid in darkness at 25 ̊C. Hairy root growth profiles in shake flask, increased periodically during 35 days of growth cycle. The cannabinoid contents produced in minor levels and remained below 2.0 μg/g dry weight. The contents of cannabinoid were analyzed by liquid chromatography and confirmed by mass spectrometry.

Cannabis sativa L. genetically transformed root based culture via Agrobacterium rhizogenes

It is an increased interest in the therapeutic potential of Cannabis sativa L. (marijuana) for treatment of multiple sclerosis and HIV neuropathy. Because of limitation in cultivation of this plant, an efficient hairy root induction system for Cannabis sativa L. was developed in the present study. Agrobacterium rhizogenes mediated transformation performed by two different co-cultivation mediums...

Root cultures of Linum spp. (section Syllinum) as a source of 6-methoxypodopyllotoxin

Linum spp. (Linaceae) has been shown to accumulate cytotoxic lignans such as podophyllotoxin and 6-methoxypodopyllotoxin. Therefore, plant cell and tissue cultures of Linum spp. may be possible alternative sources for the lignan production. In this study, we focused on the production of lignans by root cultures of Linum spp. Hairy root cultures of L. nodiflorum, L.album and L. austriacum and ro...

Analysis of choline and atropine in hairy root cultures of Cannabis sativa L. by capillary electrophoresis-electrospray mass spectrometry.

We describe a capillary zone electrophoresis method coupled to electrospray ionization (ion trap) mass spectrometry (CZE-ESI-MS) for the identification and determination of choline and atropine compounds in hairy root extracts from Cannabis sativa L. Fused-silica capillary and an alkaline solution of 20 mM ammonium acetate at pH 8.5 are used being the most suitable for the analysis of choline a...

Hydrogen peroxide Acts as a Secondary Messenger for Production of Silymarin in Ag+ Elicited Silybum marianum Hairy Root Cultures

This study was performed to investigate the effect of Ag+ as an elicitor on the activity of antioxidant enzymes, content of H2O2, accumulation of total tocopherol and antioxidant activity in hairy root cultures of Silybum marianum (L.) Gaertn. The hairy roots were treated with 2 mM Ag+. The treated and non-treated hairy roots were harvested in the same time and data on H2O2 content, DPPH, tocop...
